傳統的細胞培養過程中,並無法即時知道細胞的生長情況,為了得知細胞的即時情況必須建構一套線上即時檢測系統。
在本研究中,一個應用影像處理法結合顯微鏡構成的線上顯微光學檢測系統,將被使用在細胞培養的即時檢測上。本系統包含了細胞培養環境、顯微鏡光學平台、影像擷取系統及影像處理系統。利用影像處理的原理,搭配LabView來撰寫所需要的程式,將擷取自顯微鏡上的影像做細胞定義,使影像上屬於細胞部份的像素,能經過二值化後完全被顯示出來,並與培養液的像素作區隔。
所得的細胞像素經計算後可得整體培養皿的細胞數,然後利用血球計數器計算培養皿的細胞數為基準。經由實驗證實,線上顯微光學檢測系統可完全計算出真正的細胞數,以達到長時間細胞或組織培養期間線上即時監控細胞成長變化。
In the traditional cell culture process. It’s not readily know the growth of cells. In order to know the real-time cell must construct a set of online real-time detection system.
In this study, an application microscope with image processing method and composition of the online micro-optical detection system, will be used in cell culture on the real-time detection. The system includes a cell culture environment, optical-microscope platform, image acquisition systems and image processing system. The principle use of image processing, with LabView to write a program, will be extracted from the microscope images so cell definition images are part of the pixel cells can result after binarization completely displayed, and culture medium for the segmentation of the pixels.
The pixel of cells obtained by calculating the overall dish after the cell number, and then the use of blood cell counters calculate the number of Petri dishes as a benchmark. Confirmed by experiments, online micro-optical detection system can be fully calculated real cell number, so as to achieve long cells or tissue culture cells during the growth of online real-time monitoring of changes.
