皮膚傷口癒合過程中會產生大量自由基 ( free radicals ),導致傷口發炎,延緩傷口癒合速度。本研究利用高分子微胞 ( polymer micelles ) 包覆脂溶性維生素E 、強抗氧化劑維生素C 和保溼性極佳的γ-聚麩胺酸 ( γ-PGA ) 做為傷口癒合溶膠 ( sol )。實驗首先檢測各抗氧化劑清除超氧陰離子自由基、過氧化氫和次氯酸活性氧的能力,結果顯示維生素C、E具有超強抗氧化的能力,其中維生素C加入γ-聚麩胺酸能更有效的清除自由基,遂以此三種做為傷口癒合溶膠,以進行大白鼠全深度傷口的治療評估。在Wistar大白鼠背部開四個1×1cm2的全深度傷口,每天早晚在傷口塗抹抗氧化溶膠、溶膠、生理食鹽水和添加抗氧化物之生理食鹽水以做為對照組,十天後取下皮膚組織做組織切片,以Masson氏三色染法觀察組織和膠原蛋白生長情形,結果顯示,塗抹抗氧化物溶膠和添加抗氧化物之生理食鹽水的傷口癒合速度較快,膠原蛋白的增生也較多。
因此本研究推論,以高分子微胞包覆維生素E並添加維生素C和γ-聚麩胺酸所製做的溶膠,對傷口癒合確實具有良好的效果。
In this study, polymeric micells were encapsulating vitamin E, vitamin C, or γ-polyglutamic acid ( γ-PGA) to serve as wound healing sol. The ability to remove reactive oxygen species such as superoxide anion radicals, hydroxyl peroxide, and hyperchloric acid was evaluated. The results show that both vitamin C and E exhibited high anti-oxidation ability. The removal of free radicals was improved by combining vitamin C and γ-PGA. These 3 formulas were further evaluated with the full-thickness wound test using rats. The results show that wound healing was faster when anti-oxidant containing sol and physiological saline was applied. Thus polymeric micells ecapsultated anti-oxidant such as vitamin C, E, and γ-PGA exhibited satisfactory wound healing effect.
